蛋白體學是研究一生命體,組織或是細胞在某一生理或病理的狀態下,其整體蛋白質的量與質的變化。

本核心實驗室成立之宗旨為配合本中心計畫的研究重點『國人重要之癌症及感染症』,利用蛋白體學技術平台探究與疾病相關的關鍵蛋白質身分之鑑定、功能與其分子機轉闡明,並作為診斷、治療與基礎研究的重要科學依據。所以,本核心實驗室的主要目標為:
   1.新標定蛋白質藥物之鑑定。
   2.目標蛋白質之確定。
   3.藥物之藥理剖析。
   4.藥物之蛋白質體。
   5.比較正常與致病組織蛋白質差異性的表現。
   6.評估與追蹤藥物治療之成效。
   7.蛋白質轉譯後修飾之鑑定。
   8.與基因質體結合之策略。

中文摘要
隨著各個物種的基因體序列解碼完成,許多解決生物醫學問題之新方法陸續發展出來。因為蛋白質位居細胞功能調控的主要樞紐,蛋白體學便成為研究者高度興趣的研究課題。在後基因體時代,了解蛋白質的特性後就可連結到其對應的基因,也就是說瞭解一特定蛋白質的功能也就明瞭其對應基因的功能。
蛋白體學實為功能性基因體學的集合,旨在研究物種當中的多種蛋白質表現調控。這些研究將基因體序列和基因功能之間的關係闡明,因此世界上已經有許多蛋白體核心設施投入此方面的研究工作,國立台灣大學執台灣學術研究之牛耳,因此建立蛋白體核心設施提供蛋白體以及生物資訊方面的服務,並引進了新型的質譜儀技術作為蛋白質定性及定量方面的分析工具,包含了胜肽序列,後修飾作用,以及蛋白質之間的交互作用皆可得到更詳細且深入的瞭解。再者,核心設施並提供蛋白質分離技術,相關的生物資訊以及資料處理方面的參考。我們並不斷進行癌症及重大感染性疾病方面相關的蛋白體研究以提供此類疾病的研究基礎。透過對於疾病的徹底瞭解以及新的發現,對於研究新的治療方式和早期偵測將會有很大的助益。綜合以上所述,蛋白質核心設施即是以提供生化研究以及臨床應用方面的全方位支援,以利研究癌症,感染性疾病的發病機轉,和新藥開發。

Abstract
The completion of the genomes for a large number of organisms has begun to revolutionize experimental approaches to a variety of problems in biology and medicine. The proteome is of intense interest to investigators because proteins are the major functional components of the cell. In the post-genomic era, characterization of a protein links it directly to its corresponding gene; understanding the function of a particular protein leads directly to gene function.
Proteomics, a subset of functional genomics, is the study of the proteins expressed by an organism. These studies link genome sequence data and gene function. Many proteomics facilities have been established worldwide. As a leading academic institution in Taiwan, the facility offers a wide range of proteomics and bioinformatics services. New technologies in mass spectrometry are allowing qualification and quantitation protein expression profiles, peptide sequence analysis, post-translational modifications, and protein interactions to be studied in increasing detail. Furthermore, the facility offers consultation and support for protein separation techniques, proteome-related bioinformatics and data managements. Our ongoing projects are designed to identify all the components of cancer and infectious disease proteomes, providing a basis for functional studies. We will also address identification of functional components of selected subcellular components. The overarching goal of achieving new understanding of diseases should yield important new targets for therapeutic interpretation and markers for early detection as well. The stated mission of the proteome core facility is to provide comprehensive technological support for biomedical research, both basic and with potential clinical applications, on topics ranging from cancer and pathogenesis of infectious disease to drug design.

目標特點
1.) The discovery of proteins and genes that are associated with diseases and the determination of which of our discoveries will be useful as novel targets for drug development or as bio-markers of human disease such as cancer and infectious diseases. Proteomics also can help to identify side effects in drug candidates that might not otherwise be identified until expensive and lengthy clinical trials.
2.) The discovery of proteins in body fluids that can be used as biomarkers to assess and monitor disease. Measuring biomarkers may also lead to new diagnostic tests.
3.) Standardization of methods to separate proteins in tissue samples that improve scale, throughput and reproducibility and enable the creation of protein expression maps that can be archived for high fidelity, long-term storage and retrieval.
4.) Combined strategy with Genomics. The availability of the sequence of the human genome greatly accelerates the study of the proteome by providing the list of possible proteins in the human body, and proteomics provides clues as to which proteins and isoforms play a role in disease.
5.) Global analysis of signaling networks by proteomics.
6.) Develop a base of expertise in proteome studies and functional genomics through hiring skilled personnel and a post-doctoral training program.
7.) Support a variety of educational functions intended to disseminate information about new proteome technologies and train researchers.

本核心實驗室發展重點分兩方向:50%服務;50%研發。分別說明如下:
(I) 服務:
設置完整的蛋白質體學技術平台,包含二維電泳工作站、微量蛋白質定序、蛋白質質譜鑑定與蛋白質晶片分析,以期提供校區研究同仁高品質的蛋白質體學分析以助於其尖端研究之相關發展;本實驗室亦建立多色螢光標定系統技術,快速並正確地分析出差異性蛋白質點,以找出疾病相關蛋白質。依據服務項目之不同需求,本核心實驗室分別提供教學服務與完整操作之服務以供同仁選擇;在開授蛋白質體醫學相關教學課程及實習方面,本核心實驗室亦定期舉辦研習營,以提昇國內同仁對蛋白質體學技術之熟習與了解。
(II) 研發:
利用蛋白體學技術平台,以『國人重要之癌症及感染症』為研究重點,探究與疾病相關的關鍵蛋白質之身分鑑定、功能與其分子機轉闡明,進一步作為診斷、治療與基礎研究的重要科學依據。
